SEMIAUTOMATIC SYNTHESIS OF OLIGODEOXYNUCLEOTIDES WITH TERMINAL PHOSPHATE GROUPS BY H-PHOSPHONATE APPROACH

Abstract: Oligodeoxynucleotides (13–24 mers) were synthesized by H-phosphonate approach using a specially constructed semiautomatic synthesizer. Total synthetic cycle time was less than 6 min and average yields per coupling step were 92–97%. Up to 30 OD260 of pure products were obtained from 0.5—1.5 μmol scale synthesis after two-step HPLC purification.

To obtain 3'- or 5'-phosphorylated oligonucleotides, it is suggested to use 2-[2-(4-methoxytrityloxy)ethylsulfonyl]ethyl-H-phosphonate, which can be introduced under standard conditions of the H-phosphonate synthetic cycle and eliminated without any additional treatment.
