Synthesis and properties of 2'-deoxynucleoside 5'-phosphites labelled by tritium in the phosphite group

Abstract: Liquid-phase and solid-phase heterogeneous isotopic exchange of gaseous tritium-with hydrogen in thymidine 5'-phosphite, 2'-deoxyadenosine 5'~phosphite and 2'-deoxy— 5-oxymethyluridine 5'-phosphite has been studied. The exchange reactions are catalysed by palladium on barium sulfate. It was shown by 3H NMR and hydrolysis data that tritium is localized completely in the phosphite P—H group. The [3H]phosphites are hydrolytically stable at 20° ? within the pli range 2 to 10 and within 2 h at pH 10 and 100° C; no isotope exchange with vater was observed unler these conditions.
